Impact of Acid Ceramidase Activity on MIF-mediated migration of circulating osteoclast precursors to periodontal bone lesions in relation to aging
Description: NARRATIVE Approximately 70% of population of the aging society of the U.S. population, contrast to 40% of young and mid-age population, is affected by some form of periodontal disease leading to bone loss in the periodontium. We discovered new class of inflammatory ceramides, phosphoglycerol dihydroceramide (PGDHC) produced by Porphyromonas gingivalis, that is not only abundantly present in human periodontitis lesions, but also engaged in promotion of osteoclastogenesis. The proposed studies will investigate how age-dependent reduction of acid ceramidase (aCDase) activity that possesses host protective role by degradation of PGDHC, affects pathogenic osteoimmunological response that lead to elevated bone resorption in periodontitis lesions in elders.
