Contributions of sleep to preclinical and clinical Alzheimer's disease
Description: PROJECT NARRATIVE Identifying the specific aspects of sleep that relate to incident dementia is the first step towards the development of sleep interventions to reduce dementia risk. We propose to bring together 5 large community- based cohorts (Atherosclerosis Risk in Communities Study, Cardiovascular Health Study, Framingham Heart Study, Study of Osteoporotic Fractures, and the Osteoporotic Fractures in Men Study) with methodologically consistent polysomnographic studies and neurocognitive outcomes to comprehensively examine 134 traditional (e.g., REM and slow wave sleep percentage) and novel (e.g., EEG spectral activity, slow oscillations, and spindle density) sleep biomarkers with respect to incident Alzheimer's dementia and its cognitive and brain MRI endophenotypes in non-demented participants. This research could help us to better understand Alzheimer's disease biology and to identify new biomarkers and treatment targets.
