Novel roles for the Alzheimer's Disease (AD) risk gene, SORLA in neuroprotection in AD
Description: PROJECT NARRATIVE Our results indicate that an AD risk gene SORLA, can protect neurons from synaptic dysfunction with exposure to toxic Aβ peptides which have a role in triggering neurodegenerative onset in age-related, Aβ-dependent disorders such as Alzheimer’s disease (AD) and Down syndrome (DS). The focus of this study is to uncover how SORLA confers these neuroprotective effects, and to determine whether enhancing SORLA-associated neuroprotection can slow or reverse cognitive impairment in AD. As numerous coding mutations have been associated with SORLA in AD, it is likely that these mutations may compromise SORLA-dependent synaptic protection, thereby accelerating cognitive decline in AD.
