Consequence of senescence in muscle regeneration and degenerative myopathies
Description: Cellular senescence is a form of stress response characterized by the permanent cessation of cell division and is associated with various physiopathological processes. Specific removing of senescent cells has beneficial effects in many aging-related pathologies in mice. There is a tremendous potential in developing senotherapy for healthy aging. Yet the cause and consequence of senescence is largely unknown. The regeneration capacity of the skeletal muscle declines significantly during aging. Duchenne muscular dystrophy (DMD) is a lethal degenerative neuromuscular myopathy. Recent studies revealed a potential link between senescence and muscle regeneration. However, the mechanisms by which senescence regulates muscle regeneration and degeneration remains largely elusive. To tackle these challenges, we will develop a research program around the following two major axes: 1) Functional characterization of the senescence program on muscle regeneration, with focus on factors that control the cell cycle and senescent status in cellular populations that have a pivotal role in muscle repair 2) Characterization of the senescence program in DMD pathophysiology, with focus on strategies that can provide therapeutic benefit via senescence modulation. Overall, this project aims to systematically investigate the effect of senescence in muscle regeneration and DMD, which might provide vital information in developing novel therapeutic approaches to retard DMD progression.
