Effects of NAD restoration on neurovascular coupling in community dwelling older adults
Description: PROJECT NARRATIVE Normal cognitive function requires an adequate blood supply to support neuronal activity via neurovascular coupling, and recent evidence demonstrates that age-related cognitive decline is directly linked to reduced neuro- vascular coupling (NVC) responses. Based on the abundant literature in the field, our published preclinical results and preliminary data in humans, these series of studies are designed to assess a clinically relevant intervention to improve endothelial function and NVC responses in community dwelling older adults to prevent and, potentially to treat, age-related cognitive decline. We demonstrated that age-related decline in cellular nicotinamide adenine dinucleotide (NAD) levels is a critical mechanism that promotes impairment of cerebromicrovascular endothelial function and NVC responses, and we propose to target this mechanism via safe-to-use, as demonstrated in recent clinical trials, NAD supplement nicotinamide riboside in a double blinded, placebo controlled, parallel group study.
