Reducing the post weight-loss energy expenditure gap with orexin agonists

Description: Project Narrative Following weight loss, the new lower body weight is extremely difficult to maintain. Many studies have shown this is because individuals who lose a significant amount of body weight typically have a “Calorie Gap” post- weight loss, meaning that after weight loss, it requires fewer calories to maintain this new weight than it does for individuals at that same weight who never had obesity. A brain neurochemical, called orexin, drives calorie- burning. In this application, we will test whether peripheral administration of compounds mimicking orexin reverse this “calorie gap”, and thus prevent weight regain in animal models of obesity.
