Investigating p16 Loss in Pro-tumorigenic Metabolism
Description: Project Narrative The cell cycle regulator p16 is deleted or silenced in >30-40% of melanomas, but no current treatment strategies exist to target this patient population. In this study, we will investigate the role of p16 loss in upregulating pro- tumorigenic nucleotide metabolism by a newly-defined ATR-mTORC1 axis and evaluate inhibition of this signaling pathway in p16-low melanomas alone or in combination with current standard-of-care treatment. The results will have a direct impact on the development of novel therapeutic strategies to obtain a therapeutic response in melanoma patients with low p16 expression.
