Investigation of compartmental PDHX during cellular senescence
Description: PROJECT NARRATIVE Accumulation of senescent cells is one of the hallmarks of aging and a key contributor to tissue deterioration and organismal aging. Examining potential metabolic and epigenetic crosstalk, we identified Pyruvate Dehydrogenase Complex Component X (PDHX) as a regulator of senescence. While PDHX is predominately found in the mitochondria, we found that during senescence a nuclear isoform is upregulated, and it increases association with chromatin. The work proposed in this project will vigorously investigate the mechanistic underpinnings of compartmental PDHX in regulating metabolism and chromatin dynamics in senescence.
