Elucidation of the principles of hematopoietic stem cells in aging and overcoming aging-related diseases
Description: 本研究では、申請者がこれまでに同定した加齢造血幹細胞の機能低下の責任候補遺伝子である分子シャペロンClusterin (Clu)に着目し、その分子機序を明らかにすることで加齢に伴う造血幹細胞機能低下の原理を明らかにする。加えて、加齢関連疾患として知られる骨髄異形成症候モデルであるDNA脱メチル化酵素Tet2の機能不全マウスにおいて、Clu陽性造血幹細胞の働きを調べることで、その病態基盤の理解を試みる。これらの一連の目的を達成することで、造血幹細胞の加齢機序を理解し白血病を代表とする加齢関連疾患の克服を目指す。
