Study of neuronal regeneration using C. elegans
Description: Using mechanosensory neurons of C. elegans involving gentle touch sensation, we have previously shown that loss of either let-7 miRNA or Insulin (IIS) receptor, daf-2 help overcome the age related decline in axon regeneration and functional recovery. Similarly, swimming-related physical exercise also promotes functional recovery after axon injury through the energy sensor AMPK. Using the reporters for chemical and electrical synapses, optogenetic tools, and behavioural assays we will address how regenerating axons rewire into functional circuits in the above mentioned conditions. We will also study how the downstream targets of these pathways promote regeneration. The molecular mechanism of dendrite regeneration is poorly understood. Using PVD neuron as a model, we have found that the RAC GTPase, CED-10 that controls phagocytosis is required for dendrite regeneration. Using genetics and in vivo imaging, we will decipher the upstream and downstream pathways to CED-10 in this process. We will also perform a RNAi screen to identify novel genes controlling dendrite regeneration. Finally, we are going to set up a screening platform for accessing the efficacy of chemical compounds that can overcome regeneration failure due to aging. Overall, our work would help inform therapeutic strategies for the treatment of nervous system injuries in humans
