Understanding mechanisms of impaired immunity in the AML bone marrow

Description: PROJECT NARRATIVE Immune-based therapies have transformed the treatment of many hematologic malignancies, but such approaches have had limited success for acute myeloid leukemia (AML), an aggressive hematologic malignancy that remains fatal in nearly half of diagnosed cases. However, there is great potential for T cell- based immunotherapeutic approaches for treating AML given (1) studies demonstrating impaired T cell immunity in AML patients, (2) clinical data illustrating correlation between the bone marrow immune landscape and treatment responses to both chemotherapy and immunotherapy, and (3) the curative potential of allogeneic hematopoietic cell transplantation for AML arising from the immune-mediated graft-versus-leukemia activity of donor T cells. The goal of this study is to perform deep analysis of T cells in parallel with surrounding populations in the bone marrow, the site where AML emerges, to elucidate key mechanisms driving T cell functional responses to AML longitudinally, and to identify immunologic targets for potential therapeutic intervention.
