Gene-lifestyle interactions behind subclinical cardiovascular disease - explorative analyses based on genome-wide data
Description: It is generally recognized that cardiovascular disease etiology involves complex interplay of genetic susceptibility and environmental exposures. However, still little is known about such interactions. The overall aim of this project is to identify risk groups in the population, genetically susceptible to the exposure to specific lifestyle factors. Specifically, this project aims at exploring about 200,000 genetic variants, selected with an unbiased approach, which may modify the association between lifestyle related cardiovascular risk factors and the progression of subclinical cardiovascular disease. The life stylefactors to be considered are cigarette smoking, physical inactivity and main type of dietary fat consumed. Sublinical cardiovascular disese will be assessed using measurements of carotis intima media thickness. The participants of the IMPROVE study (n=3,691) will form basis for the analyses. C-IMT was measured at baseline investigations, in 2004-2005, and after 10 years of follow-up. The C-IMT progression over time will be calculated and fast C-IMT progression will be defined as progression rate in the upper quartile of the distribution. Associations between genetic variants and fast C-IMT progression will be modelled using logistic regression. Odds ratios will be adjusted for age and sex. To obtain personalized cardiovascular disease prevention there is a need to identify gene-lifestyle interactions.
