Comprehensive characterization of histone epigenetic marks

Description: Histone post-translational modifications (PTMs) are essential epigenetic regulators of gene expression. As the imbalance in the modification status often leads to the disease formation, the development of methods for deeper histone characterization is of critical importance. Currently, approaches based on mass spectrometry (MS) have been applied as the first choice for PTMs analysis. The determination of histone marks is addressed mostly by shotgun proteomic analysis in combination with chemical derivatization of histone lysine residues. However, those analyses are challenging, mainly due to multiple histone variants and their modification diversity. In the project, we will develop methodology for histone characterization in both mammal and plant systems. We will particularly focus on improvement of chromatographic properties of histone peptides using novel derivatization agents, digestion using alternative proteases and use of advanced MS acquisition methods. Established protocols will be combined to a net workflow enabling paralel characterization of multiple types of histone PTMs.
