Peptide-Directed NMDA Receptor Modulation: Implications for Food Motivation, Cognition, and Neuroplasticity
Description: Obesity is a major public health concern, associated with increased risk of neurodegenerative diseases. Glucagon-like peptide-1 receptor agonists (GLP-1RAs) have shown great promise in promoting weight loss and improving metabolic health, yet sustaining these effects remains challenging. Interestingly, many genes associated with adiposity are linked to NMDA receptor (NMDAR)-driven neuroplasticity, highlighting a potential neurobiological component in metabolic regulation. While NMDAR antagonists are primarily used in the treatment of neurodegenerative disorders, they also influence appetite and synaptic plasticity. This project explores the therapeutic potential of GLP-1 directed-NMDAR antagonism in modulating food-motivated behavior, cognitive function, and synaptic plasticity. Using rodent models, we will examine how GLP1agonist/NMDAR antagonist conjugates affect feeding behavior, learning, and reward processing under both normal and metabolically challenged conditions. Additionally, we will investigate neuroinflammatory mechanisms and sex-specific responses to treatment. By bridging metabolic and neurobiological perspectives, this research aims to uncover novel pharmacological strategies for obesity and cognitive disorders, paving the way for more effective and personalized treatments.
