Assessing cellular aging in old and rejuvenated neurons from Alzheimer patients
Description: PROJECT NARRATIVE The project by Drs. Gage, Mertens and colleagues aims at deciphering the roles of age-related aberrant PKM2 and neuronal hypo-maturity in Alzheimer’s disease (AD) using patient-specific induced neurons (iNs). The team will study the spatial relationship between neuronal PKM2 and markers for neuronal senescence, neurodegeneration and neuroinflammation in brain sections from AD patients, and employ functional and unbiased multi-omic approaches to study the functional impact of engineered PKM2/1 imbalance in vital human iNs. They will also leverage insights from the cancer field and explore chemical PKM modulators in AD patient iNs to develop novel approaches to treat AD, and they will assess the impact of aberrant metabolic states of aged human neurons in the context of neuron-astrocyte coupling in a three- dimensional multicellular model based on age-equivalent iNs. 1
