Mechanism of action and function of novel secosteroid 20(OH)D3 in the skin
Description: During this renewal, using the best available in vivo models and state of the art methodology, we will define the protective role of novel CYP11A1-derived vitamin D3 hydroxymetabolites against UVB induced epidermal pathology including photocarcinogenesis. Through highly mechanistic studies we will define the nature of their anti-oxidative and DNA protective effects and mechanisms by which they regulate keratinocyte differentiation program with a focus on specific nuclear receptors (NRs). We will also investigate mechanistic links between the candidate NRs and downstream regulatory pathways.
