Aberrant Micro-managing of the Airway Epithelial Transcriptome in HIV-associated COPD
Description: PROJECT NARRATIVE: COPD is a multifactorial pathology and PLWH demonstrate and higher incidence of COPD even when adjusted for the smoking status. HIV tat and TGF-β signaling (upregulated by Tat, cigarette smoek and in COPD), dysregulate the airway microRNAome and this can have downstream effects on the transcriptome and signaling pathways leading to pathological hallmarks of COPD. Understanding the mechanism of miRNA dysregulation and its downstream effects can identify targets for intervention, and silencing HIV transcription by CRISPR based inactivation of cellular cofactor CyclinT1 only in HIV-infected cells can decrease the Tat burden in the airways and improve outcomes in HIV associated COPD.
