Tissue systems biology of immune dysregulation in aging by single cell spatial metabolomics
Description: PROJECT NARRATIVE The proposed research is relevant to public health because it will build a foundational metabolic understanding of tonsils and lymph nodes, common immune organs playing a pivotal role in immunity, and their structure and functional alterations during aging. This proposal leverages cutting-edge spatial metabolite and protein profiling technology to define chemical regulators and defects in controlling a key cell type, B cell, in human tissues from diverse aged donors while simultaneously testing physiologically relevant differences between anatomically distinct T cells and B cell subtypes with unique functional changes in aging. Thus, the proposed research is relevant to the NIA’s mission of obtaining new knowledge on how the immune system is dysregulated in the elderly compared to young individuals through tissue-level molecular studies.
