DC exosome therapy to resolve inflammatory bone loss and oral infection

Description: Project Narrative Periodontitis (PD) is one of the most common inflammatory diseases in the U.S, contributing to increased risk of other diseases with more serious mortality and morbidity profiles, but as yet there is no effective immunotherapy for PD. Understanding how gingival immune cells contribute to inflammatory bone loss and how to “reprogram” them to preserve bone, using dendritic cell-derived exogenously developed exosomes, is the goal of this proposal. Our first exosome prototype, enriched with TGF and IL-10, fulfills this novel premise in mice, but the precise mechanisms involved, and the direct relevance to endogenous exosomes from humans with PD need to be established, as outlined in these aims.
