Molecular clock dysfunction in lung cellular senescence by environmental tobacco smoke
Description: PROJECT NARRATIVE Molecular clock dysfunction is associated with the systemic effects of lung irritants like environmental tobacco smoke (ETS) exposure, however, the impact of clock dysfunction on ETS-mediated cellular senescence (irreversible growth arrest) and premature lung aging is not known. We will determine the mechanisms by which the molecular clock regulates stress-induced cellular senescence and inflammation, and whether pharmacological augmentation of REV-ERB and SIRT1 (anti-aging) activity can attenuate the stress-induced senescence caused by chronic ETS exposure. This will have great translational potential in the development of pharmacological approaches to ameliorate premature lung aging in airway disorders by adverse effects of ETS.
