Regulation of ovine follicle-stimulating hormone beta-chain mRNA by 17 beta-estradiol in vivo and in vitro.

Abstract: 17p-Estradiol (E2) administration causes a dramatic decrease in levels of follicle-stimulating hormone (FSH) in the sera of castrate male sheep and in the secretion of FSH from primary cultures of ovine pituitary cells. The possibility that this control occurs by changes in specific mRNA levels was investigated.FSH fl-chain mRNA activity, as measured by cell-free translation of pituitary polyadenylated RNA in a wheat germ system, followed by specific immunoextraction of pre-FSH#?, was found to parallel closely the decrease in serum FSH levels of castrate male sheep caused by daily Ez administration (1 mg/day).Serum FSH and pituitary FSHb mRNA activity levels were reduced about 75% after 3 days and 85-908 after 6 days of Ez treatment.Other variables, such as pituitary size and mRNA yields, did not vary significantly between treatment groups.Similar but more rapid effects were obtained using Ez in primary cultures of pituitary cells.FSHP &NA activity and FSH secretion rates were both reduced approximately 50% after 12 h of incubation with 10 n M Ez and were inhibited 80 and 90% after 24 and 48 h, respectively. The high correlation of FSHP mRNA activity and FSH secretion was also observed over a wide range of Ez concentrations. The dose of Ez required for 50% inhibition in a 48-h incubation was approximately 30 PM (8 pg/ml), which is within the physiological range of serum E2 concentrations in normal ewes.By translating mixtures of pituitary mRNAs from control and Ez-treated sheep, it was found that E2 did not induce RNA(s) that inhibited the translation of ovine FSHB mRNA.These results provide evidence that EZ decreases the production of FSH by suppressing the levels of mRNA coding for the hormone-specific p-subunit.17p-Estradiol plays an inhibitory role in the synthesis and secretion of follicle-stimulating hormone in several species (1).For example, physiological levels of E21 rapidly and reversibly
