The Mechanism of Action of Ethanolamine Ammonia-Lyase, a B12-dependent Enzyme
Year: 1971
Type: article
Abstract: When acetaldehyde and ammonium ion are incubated with ethanolamine ammonia-lyase and coenzyme B12, two processes occur: (a) transfer of hydrogen from the C-5' of the coenzyme to acetaldehyde and (b) dissociation of the coenzyme at the carbon-cobalt bond with the formation of 5'-deoxyadenosine and enzyme-bound hydroxocobalamin. When NH4+ is omitted, no tritium transfer to acetaldehyde occurs, but the dissociation of the carbon-cobalt bond is accelerated. From the results it was concluded that 1-aminoethanol is an intermediate in the reaction.
