Abstract: Bone tissue engineering has shown limited success, mostly owing by lack of vascularization leading to poor graft integration. Some strategies can enhance vascularization in engineered bone construct. One of them result on the observation that endothelial cells (ECs) cocultured with osteoprogenitor cells can formed prevascular structures in vitro which can, after implantation, anastomose spontaneously to the ingrowing vasculature of host. The cross-talk between osteoblasts and ECs into the coculture creates a suitable environment to promote angiogenesis, to induce formation of a perivascular network and to stimulate osteoblastic differentiation. Here, we assumed that using cells derived from the same stem cell source can promote cell-cell interactions and osteoblastic differentiation. The aim of this study was to investigate if SCAPs can differentiate toward functional endothelial cells (SCAPs-E) and express endothelial markers. W! e studied the immunophenotype of SCAPs by flow cytometric analysis. The endothelial markers of SCAPs-E ( CD31 and vWf) were assayed by histochemical methods and real-time PCR. We demonstrated that SCAPs expressed a heterogeneous panel of makers associated with stemness. Moreover SCAPs-E expressed von Willebrand Factor (vWf) but not CD31. Also, our study focuses on the ability to seed SCAPs and endothelial cells by laser assisted bioprinting in order to control their organization on collagen. Cells viability was validated by a live/dead test.
