Vaccinia Virus Vectors

Year: 1992
Type: book-chapter
Abstract: Publisher Summary This chapter discusses vaccinia virus vectors. With the elimination of a deadly disease and the eradication of the offending pathogen, the smallpox vaccine holds a unique place in preventive medicine. The characteristics of the vaccine that contributed to its effectiveness against smallpox, including low cost, heat stability, simple methods of administration, visible evidence of successful vaccination, and a long duration of immunity following a single dose have inspired efforts to develop vaccinia virus into a vector that can be used for the prevention of other infectious diseases. With the aid of recombinant DNA technology, vaccinia virus can be engineered to express one or more genes derived from virtually any microorganism. Such recombinant viruses retain infectivity and induce humoral and cell- mediated immune responses upon injection into susceptible hosts. These vectors have proven to be extraordinarily useful reagents for vaccine-related research and are undergoing evaluation as candidate vaccines.
