Abstract: Trials that are both very large and simple are often needed to provide reliable answers to important clinical questions. Large, simple trials can demonstrate or exclude clinically meaningful, moderate treatment effects. The large sample size is necessary to overcome moderate biases and moderate random errors. The simple structure and conduct is necessary to make such trials feasible and practical. The various characteristics of large, simple trials will be illustrated with published examples.
