Differential labeling and identification of the cysteine-containing tryptic peptides of catalytic subunit from porcine heart cAMP-dependent protein kinase.

Type: article
Abstract: The cysteine-containing tryptic peptides from the catalytic subunit of CAMP-dependent protein kinase were isolated and characterized with the aid of the sulfhydryl specific reagent iodoacetic acid.When the catalytic subunit was reacted with ['4C)iodoacetic acid, cleaved with trypsin, and chromatographed on Sephadex G-50, three distinct components of radioactivity (C-1, C-2, and C-3) were identified.These components were further purified using a combination of high performance liquid chromatography and paper chromatography.Subsequent analysis revealed that only two of these components ((3-2 and (2-3) contained carboxymethylcysteine.The sequences of these two peptides are as follows: (C-2) -Cys-Gly-Lys-Glu-Phe-Ser-Glu-Phe;
