Fine mapping of the rrnE, purHD, and hydGH operons on the Escherichia coli chromosome

Year: 1991
Type: article
Abstract: The nucleotide sequence of the hydGH operon was re-ported recently by Stoker et al. (6); the operon was observed to be part of a multicomponent system regulating hydroge-nase activity in Escherichia coli. However, the precise chromosomal location, or linkage to adjacent genes, was not determined (2, 4). Similarly, the nucleotide sequence of the purHD operon was reported by Aiba and Mizobuchi (1), and its position on the chromosome was approximated at 90 min or between 3496 and 3500 kb (2, 4, 5). PurH and PurD are involved in the de novo synthesis of purine nucleotides in E. coli. and relative direction of transcription of all three operons vis-a-vis the chromosome. By joining the overlapping sequences of the independently cloned DNA fragments encoding the rrnE promoter (3), hydGH operon (6), and purHD operon (1), it was possible to define the position of the hydGH operon on the E. coli physical map. Figure 1 illustrates the physical relationship of the three gene clusters and shows the regions of overlap between the rrnE, hydGH, and purHD fragments. The region of overlap between the hydGH and purHD fragments consists of 362 bp and the residue identity was 98.1%, (physled map b_ton) 'Fagnu oveueprgon (59 bP,983%Id_u): *. |PU (301-1890 bp), 301 gi
