480. Gene Transfer to Gut K Cells for Physiologic Insulin Replacement in Diabetes
Year: 2006
Type: article
Abstract: We have previously demonstrated the feasibility of using gut K- cells as surrogate cells for insulin production. Transgenic mice expressing human insulin specifically in K-cells remained normoglycemic and glucose tolerant following destruction of their pancreatic |[beta]|-cells. However, a vector system is needed to transfer effective amount of the therapeutic insulin gene to duodenal epithelial cells in vivo. Previous efforts using viral and non-viral gene delivery vector resulted in some success. However, persistent transgene expression and detectable circulating proteins were not observed. Accordingly, this study is to identify a suitable viral vector and an optimal delivery methodology to achieve long-term gene transfer to the duodenum.
