RNA transfected cord blood-derived dendritic cells generate anti-leukemic cytotoxic T lymphocytes

Abstract: Acute Lymphocytic Leukaemia (ALL) patients who relapse after haematopoietic stem cell transplantation have few therapeutic options. An immunotherapeutic approach that enhances the graft versus leukemia effect may improve their survival. We postulate that cord blood (CB) CD34+-derived dendritic cells (DC) will be ideal source of DC for CB transplant recipients. In addition, we propose using RNA as a source of antigen to induce T cell responses to poorly immunogenic tumors such as ALL. CD34+ cells can expand and differentiate into DC and function as professional antigen presenting cells.
