Abstract: The paper by Zielonka et al. (1Zielonka J. Sikora A. Joseph J. Kalyanaraman B. J. Biol. Chem. 2010; 285: 14210-14216Abstract Full Text Full Text PDF PubMed Scopus (163) Google Scholar) demonstrates a plateau of peroxynitrite with variable flux ratios of NO and superoxide using a probe (coumarin-7- boronic acid (CBA) that reacts directly with peroxynitrite. This study provides an elegant verification that peroxynitrite is the initial product, even though oxidative and nitration chemistry often times exhibits a bell-shaped behavior (1Zielonka J. Sikora A. Joseph J. Kalyanaraman B. J. Biol. Chem. 2010; 285: 14210-14216Abstract Full Text Full Text PDF PubMed Scopus (163) Google Scholar). However, the authors state that their results are contrary to a “bell-shaped curve” phenomenon associated with oxidation by ONOO− generated from NO and O2−. precursors. Past studies have established that 1) rhodamine is the primary product of dihydrorhodamine (DHR) oxidation by NO/O2−. (2Jourd'heuil D. Jourd'heuil F.L. Kutchukian P.S. Musah R.A. Wink D.A. Grisham M.B. J. Biol. Chem. 2001; 276: 28799-28805Abstract Full Text Full Text PDF PubMed Scopus (194) Google Scholar, 3Jourd'heuil D. Miranda K.M. Kim S.M. Espey M.G. Vodovotz Y. Laroux S. Mai C.T. Miles A.M. Grisham M.B. Wink D.A. Arch. Biochem. Biophys. 1999; 365: 92-100Crossref PubMed Scopus (65) Google Scholar); 2) the bell-shaped behavior of NO/superoxide fluxes has been established for other common detectors such as NADH (2Jourd'heuil D. Jourd'heuil F.L. Kutchukian P.S. Musah R.A. Wink D.A. Grisham M.B. J. Biol. Chem. 2001; 276: 28799-28805Abstract Full Text Full Text PDF PubMed Scopus (194) Google Scholar); and 3) tyrosine nitration and oxidation products from NO/O2−. fluxes can be all accounted for by the effect of excess NO or O2−. (4Pfeiffer S. Mayer B. J. Biol. Chem. 1998; 273: 27280-27285Abstract Full Text Full Text PDF PubMed Scopus (159) Google Scholar). Since CBA presumably reacts quickly with ONOO− prior to and faster than subsequent reaction with other species in solution (i.e. excess precursor), it is expected that CBA will not exhibit the bell-shaped curve reported for other reactants, which react with the reactive species formed from ONOO− decomposition and not ONOO− itself. Therefore the boron coumarin complex provides a complementary tool for direct detection of ONOO−, while DHR assesses the oxidative chemistry of the reactive intermediates after the rate-limiting step of ONOO− decomposition by either H+ or CO2.
