MASTering Treg Function to Promote Tolerance

Year: 2009
Type: editorial
Abstract: Rejection or tolerance of allografts depends on the activity of different types of immunoregulatory cells. The best characterized of these are CD4+ T regulatory cells (Tregs) that express the Foxp3 transcription factor. It is now well established that adoptive cell therapy with antigen-specific Foxp3+ Tregs promotes tolerance, whereas removal of these cells causes graft rejection in multiple experimental models of allograft tolerance. Although much progress has been made toward the induction of alloantigen-specific Tregs in vitro or in vivo, little is known about how Tregs mediate indefinite graft survival in this complex cellular environment, and even less is known about what can cause this process to fail.
