Abstract: Alemtuzumab, a humanized monoclonal antibody, depletes CD52+ circulating leukocytes. In humans, this includes most mononuclear leukocytes. Exciting results from 2 phase III trials established the superiority of alemtuzumab in reducing multiple sclerosis (MS) relapse rate compared with high-dose interferon-β.1 Increased mild to moderate infections occurred in both trials in the alemtuzumab compared to interferon-β arms. Also noteworthy, secondary autoimmune diseases develop in a sizeable proportion of alemtuzumab-treated patients.
