Abstract: Current evidence indicates a role for Notch signaling in regulating cell-fate decisions of hematopoietic precursors. Earlier work performed in our lab demonstrated enhanced self-renewal of repopulating cells due to retrovirus-mediated expression of a constitutively active form of Notch1. Subsequent studies demonstrated that activation of endogenous Notch receptors by exogenously presented Notch ligand was able to expand precursor cell numbers from human and murine non-mutant stem cells, including those with short-term and, in the case of human cells, possibly long-term repopulating ability.
