Cover Picture: 2,3‐Dihydro‐1‐Benzofuran Derivatives as a Series of Potent Selective Cannabinoid Receptor 2 Agonists: Design, Synthesis, and Binding Mode Prediction through Ligand‐Steered Modeling (ChemMedChem 10/2009)

Abstract: The cover picture shows the cannabinoid receptor 2 (CB2) complexed with compound MDA104. A putative hydrogen bond between S3.31 and MDA104 is represented as yellow dashes. Recently, the CB2 receptor has emerged as a new target for the treatment of neuropathic pain. Neuropathic pain is caused by lesions in the central or peripheral nervous system and is characterized by severe, persistent pain that is refractory to traditional analgesics. We designed a novel series of 2,3-dihydro-1-benzofuran derivatives bearing an asymmetric carbon that behave as CB2 agonists. Binding mode prediction through ligand-steered modeling was used. Separation of the enantiomers and assignment of their configuration led to the identification of the S-enantiomer as the eutomer. The figure was prepared using Pymol (www.pymol.org).
