Abstract: Parkinson’s disease (PD) is a neurodegenerative disorder that affect mainly aged people. Lesions induced by the 6-hydroxydopamine (6-OHDA) and 1-methyl4phenyl-1,2,3,6-tetrahydropyridine (MPTP) has been successfully used as rat models of PD. MPTP and 6-OHDA models are among the most used models to study PD. Previous studies showed that the MPTP model is useful to study cognitive deficits of PD, however it is not know if this model is valid to study motor impairments. The unilateral lesion of the rat SNc with 6-OHDA is a model very used to study motor impairments. In the present study we compare the differences between unilateral and bilateral lesions of the SNc on motor and working memory tests. The effect of dopaminergic agonists, nicotine and caffeine were tested in MPTP or 6-OHDA unilaterally-lesioned rats. Adult male Wistar rats were lesioned by the infusion of MPTP or 6-OHDA into the substantia nigra pars compacta (SNc) or the medial forebrain bundle (FPM) and submitted to the open field, water maze, and turning behavior tests. Also, post mortem analisis of the striatal dopamine and SNc histology were analyse. The bilateral of left lesion of the SNc impaired working memory. Both MPTP and 6-OHDA unilaterally-lesioned rats presented turning behavior when challenged with dopaminergic, adenosinergic and nicotinic drugs. The depletion of striatal dopamine is conspicuous in the lesioned side, as well as the SNc dopaminergic cels.
