Assessing Sleeping Beauty Transpositions for T-Cell Immunotherapy by Supercomputer-Based High-Throughput Profiling of Integration Events
Year: 2012
Type: article
Abstract: Three first-in-human clinical trials using Sleeping Beauty (SB) transposition for immunotherapy of T cells expressing a CD19-specific chimeric antigen receptor (CAR) have received institutional and federal regulatory approvals and are under way at the University of Texas MD Anderson Cancer Center. To understand potential risks involved in using non-viral SB transposition to genetically modify clinical-grade T cells, we employed high-throughput sequencing (Illumina) and profiling (BlueBioU supercomputer at Rice University) of the SB-mediated CAR insertion sites in T cells.
