Abstract: instillation of saline.Group 2-6 using cystitis model were constructed by intraperitoneal cyclophosphamide combined with intravesical protamine/lipopolysaccharide.In group 3 and 6, hyaluronic acid (HA) or anti-ICAM-1 antibody (AIA) were instilled intrvesically respectively. In group 4 and 5, celecoxib or aprepitant were introduced to rats by gavage respectively.Bladder samples were harvested for histological evaluation including inflammation grade, mast cell counts and measurement of purinergic receptors (P2X3 and P2Y2), PGE2 and E-series prostaglandin receptors (EP1 and EP2), tumor necrosis factor alpha (TNF-a), ICAM-1 and neurokinin-1 receptor (NK1R).RESULTS: Significant increase of bladder inflammation grade and mast cell counts were observed in cystitis model, accompanied with higher expression of P2X3/P2Y2, PGE2 and EP1/EP2, TNF-a, NK1R and ICAM-1.AIA significantly decreased inflammation grade and mast cell counts, while other treatments had weaker or no effect.Aprepitant attenuated expressions of all cytokines and receptors except P2X3 to some degree.AIA could inhibite the elevation of all cytokines and receptors significantly stronger than any other drugs including aprepitant.Furthermore, the expressions of all involved cytokines and receptors in rat models receiving AIA became no difference to normal control.CONCLUSIONS: Compared with HA, celecoxib or aprepitant, anti-ICAM-1 antibody showed remarkably superior effects in attenuating the severity of non-bacterial cystitis in rat model representing IC.ICAM-1 probably plays a critical role in the pathophysiological of PBS/IC and should be further investigated as a new therapeutic target.
