Abstract: Objective To investigate the change and its clinical significance of mitochondrial function in skeletal muscle in patients with amyotrophic lateral sclerosis(ALS). Methods The condition of 80 ALS patients was evaluated with ALS functional rating scale(FRS),and skeletal muscle biopsy was done. The percentage of ragged red fiber(RRF) and activity of cytochrome C oxidase(COX) of muscle specimen were detected by modified gomori trichrome(MGT) and mitochondrial enzymes COX stain. The correlation between percentage of RRF,activity of COX and FRS scores were analyzed respectively. Results In this group,RRF in muscle was found out in 13 cases(16.3%),including 9 cases with 1% RRF,3 cases with 4% RRF and 1 case with 4% RRF.The muscle COX active was deficiency in 21 cases(26.3%),6 cases with complete deficiency and 15 cases with partial deficiency. The correlations between percentage of RRF,COX active deficiency and FRS score were negative(r=-0.234,P0.05;r=-0.285,P0.01). Conclusion The mitochondrial function in skeletal muscle in ALS patients is abnormal;and which more obvious, the heavier the condition of the patient.
