Abstract: Manual 20kb library construction of PacBio libraries requires careful handling of the DNA throughout the protocol. There are many small volume steps and clean ups which need to be accurate and reproducible. At TGAC we do a lot of small microbe PacBio only sequencing that tend to only require 1 or 2 SMRT cells. The bottleneck with these projects is generating the libraries. Typical final library yields can be as low as 20% with only 5ug input. Here at TGAC, we are working on several different liquid handling systems to automate the large insert library construction and present our progress.
