Myeloablative Flu/Bu Conditioning Provides Similar Overall Survival and Lower Non-Relapse Mortality Compared to Traditional Bu/Cy or Cy/TBI in Allogeneic Transplantation

Year: 2017
Type: article
Abstract: Myeloablative conditioning (MAC) is used in allogeneic hematopoietic stem-cell transplantation (HSCT) to maximize disease control. Common regimens include cyclophosphamide/busulfan (BuCy) and cyclophosphamide with total body irradiation (CyTBI). A non-randomized prospective CIBMTR study showed that overall survival (OS) was marginally better with Bu-based rather than TBI-based regimens. However, an EBMT retrospective registry analysis showed similar leukemia free survival using CyBuor CyTBI, while acute and chronic graft-versus-host-disease (GVHD) were greater in the TBI arm.
