Ropinirole treatment resulting in recovery of sensorimotor gating induces ΔFosB in mouse nucleus accumbens neurons that co-express D1 and D3 dopamine receptors

Abstract: Background : Acute treatment with the D 2 -like dopamine receptor agonist, ropinirole, disrupts sensorimotor gating measured using prepulse inhibition of the acoustic startle response (PPI). We showed previously that repeated ropinirole produced long-lasting recovery of PPI, mediated by CREB phosphorylation in nucleus accumbens (NAc) neurons (Berger et al, Biol. Psychiatry , 2011). The present study explored whether, and in which neurons, the long-lasting transcription factor ΔFosB was expressed after repeated ropinirole treatment. Conclusions : NAc neurons expressing D1R and D3R could contain heterodimeric receptors whose repeated activation by ropinirole drives D1R-coupled signaling. This could underlie increased phosphoCREB and ΔFosB expression in these NAc neurons, which may be required for PPI recovery.
