Abstract: The pituitary gland controls the entire endocrine system and is responsible for the production and regulation of many critical hormones including GH, ACTH, LH, FSH, TSH, and prolactin. Pituitary tumors are benign growths that mainly remain in the pituitary gland, which may cause variety of symptoms including vision loss and infertility. Metabolome of pituitary tumors are largely unknown. We are reporting ex vivo NMR spectroscopy based metabolomics of two major subtypes of pituitary tumors. Pituitary tumors [LH/FSH = 3; Prolactinoma = 3] were collected from patients undergoing transsphenoidal hypophysectomy, at the Houston Methodist Hospital and were immediately snap-frozen. Tissue samples were extracted in methanol:chloroform (2:1) solvent-mixture. Lyophilized powders from methanol-layer was reconstituted in D2O containing 1.0 mM DSS (internal standard for quantification of metablites), and the residues from chloroform-layer was redissolved in CDCl3 containing 1% TMS. 1H NMR data were collected on a Bruker 600 MHz spectrometer. Pituitary tumor tissue showed the presence of major CNS metabolites: N-acetyl aspartate (NAA), glutamate, glutamine, myo-inositol (mI), and aspartate. Phosphoethanolamine (PE) was predominantly observed in pituitary tumors. Quantitative comparison of various metabolites between these two types of pituitary tumors showed decreased levels of NAA, mI, and PE and elevated levels of glutamate, glutamine, and aspartate in prolactin secreting tumors. Lipid profiles of the above two groups were comparable to each other. Currently, we are recruiting more number of patients to perform a detailed metabolomics analysis of pituitary tumors of various clinical subgroups. The metabolism in pituitary gland was altered due to tumorigenesis. Determination of pituitary metabolome in different clinical types of tumors will help towards better clinical characterization of these tumors. Further, metabolic pathways associated with key metabolites identified in altered pituitary metabolism will be used to develop molecular targets for designing new therapies for treating pituitary tumors.
