Abstract: inhibition of MCAO-OAB (p<O.Ol).Baclofen (3 mg/kg) but not diazepam (3 mg/kg) also significantly inhibited MCAO-OAB.The inhibitory effect of both gabapentin and baclofen was significantly blocked by CGP55845 (2 mg/kg).(2) CA-OAB mice.VI was reduced 44% from 7.8 ± 1.7 to 3.6 ± 1.3 min at 60 min of CA infusion.VP did not change significantly.Gabapentin (10-100 mg/kg) dosedependently inhibited CA-OAB (p < O.O I).Baclofen (10 mglkg) but not diazepam (10 mg/kg) also produced inhibition of CA-OAB.CGP55845 (2 mg/kg) significantly blocked the effect of gabapentin ( 100 mglkg) and baclofen ( 10 mg/kg) on CA-OAB.CONCLUSIONS: Like the GABA8 receptor agonist baclofen, gabapentin produced inhibitory effects on MCAO-OAB and CA-OAB in conscious rodent models and the effects were blocked by a GABA8 receptor antagonist. These results suggest that gabapentin may act either directly on the GABA8 receptor or via this downstream mechanism to produce inhibitory effects on micturition hyperreflexia.
