Detection of somatic RAS mutations in circulating tumor DNA from metastatic colorectal cancer patients: are we ready for clinical use?
Year: 2018
Type: article
Abstract: Assessment for somatic sequence alterations in exons 2, 3, 4 of KRAS and NRAS genes (expanded RAS) from tumor tissue is mandatory before initiating anti-EGFR therapy in metastatic colorectal cancer patients [1]. Although analysis for mutations is most commonly carried out on archival formalin-fixed paraffin-embedded (FFPE) tissue, this procedure is limited by sample quality, lack of methodological standardization and unreliable turnaround time. Moreover, the use of single tissue biopsy may underestimate tumor heterogeneity and will not reliably detect mutations present below 2%–3% of the sample (i.e.
