Abstract: INTRODUCTION AND OBJECTIVE: Estrogen is implicated in the pathogenesis of benign prostatic hyperplasia (BPH) as well as epithelialstromal interactions. We characterized the effect of estrogen on the expression of smooth muscle cell (SMC)-specific markers in prostatic stromal cells (PrSCs) through regulation of benign prostatic epithelial cells in a paracrine pathway.METHODS: Prostate epithelial cell line BPH-1 and PrSCs were routinely cultured. When cells grew to 30% confluence, the medium was changed into phenol red free DMEM/F12 without serum. After 24h the BPH-1 cells were treated with or without estradiol (10-6 M).Conditioned media (CM) were harvested 72h later and their effects on the expression of SMC-specific markers in PrSCs, namely smoothelin (SMTN) and smooth muscle myosin heavy chain (SMMHC), were tested.Real-time RT-PCR and western blotting were performed to determine SMTN mRNA and SMMHC protein expression, respectively. The expression of transforming growth factor 131 (TGFI31) was analyzed by real-time RT-PCR and ELISA.TGFI31 neutralizing antibody was used to characterize the effects of TGF131 in BPH-1 CM on PrSCs.RESULTS: SMTN mRNA and SMMHC protein expressions in PrSCs increased after treatment with CM from BPH-1.CM collected from BPH-1 without estradiol stimulation increased SMTN mRNA level approximately 1.8-fold after 24h and SMMHC protein level about 2 folds after 48h.The CM from BPH-1 treated with estradiol increased smoothelin mRNA and SMMHC protein expression more effectively, which were about 2.7-fold and 2.5-fold compared with control group, respectively.RT-PCR shows that estradiol increased TGF131 mRNA in BPH-1 approximately 1.75-fold after 24hr.ELISA assay confirmed this data. The concentrations ofTGF131 in BPH-1 CM treated with 0 and 10. 6 M estradiol are (176.2±7.9)pg/10 4 cells and (266.2±9.2) pg/10 4 cells, respectively. After neutralizing antibody to TGF131 was added at a final concentration of 1 IJg/ml, CM collected from BPH-1 cells treated with or without estradiol failed to stimulate SMTN and SMMHC expression in PrSCs.CONCLUSIONS: Estradiol may promote PrSCs differentiation through epithelial-stromal interactions. This study suggested that prostatic epithelial cell line BPH-1 enhances the expression of SMC-specific markers of PrSCs through the production of TGF131 in a paracrine mechanism. This effect is further augmented after stimulating BPH-1 with estradiol.
