Abstract: prostatic hyperplasia. However the mechanism responsible for the prostatic phenotype is unknown. To determine whether restoration of leptin levels normalizes proinflammatory status associated with MetS and modulates the prostate phenotype, we evaluated ob/ob mice across a range of criteria including prostate morphology and gene expression profile.METHODS: Leptin was delivered using a subcutaneous osmotic mini-pump.RESULTS: Body and liver weights of leptin-treated ob/ob mice were significantly lower than non-treated ob/ob mice, but still heavier than control C57BL/6J mice.A glucose tolerance test revealed significantly shorter recoveries in leptin-treated versus non-treated ob/ob mice.Ventral prostate glands in non-treated ob/ob mice exhibited increased epithelial hyperplasia compared to lean C57BL/6J mice. The proportion of hyperplastic prostate glands in leptin-treated ob/ob mice was significantly reduced by leptin treatment.Microarray gene ontology enrichment analysis of ob/ob mouse prostate tissue revealed that innate immune responses, especially interferon signaling, were highly enriched in non-treated ob/ob mice.Cytokine array analysis revealed that CCL6 and insulin-like growth factor binding protein family were upregulated in non-treated ob/ob prostates.Microarray gene ontology enrichment analysis of human BPH tissue revealed that cytokine activity and interferon signaling were also highly enriched in obese patients.CONCLUSIONS: These data demonstrate the feasibility of using the ob/ob mouse as a model in which weight can be manipulated to directly investigate the relationship between obesity and prostatic hyperplasia.
