Abstract: The cover feature picture shows an optical illusion that can be viewed either as a white vase or the outline of two faces. By analogy, the nitrileimine–alkene cycloaddition, well known for its use in biorthogonal protein labeling, can also enable the fluorescent detection of endogenous molecules such as the oncometabolite fumarate, which accumulates in the hereditary cancer syndrome HLRCC. Applying this method to profile the activity of tricarboxylic acid cycle enzymes allowed the facile differentiation of healthy tissue and tumor in HLRCC patient samples, and enabled proof-of-concept experiments to establish the feasibility of oncometabolite imaging.
