P1‐466: Diminished Practice Effects on a Monthly Computerized and Home‐administered Face‐name Associative Memory Exam Are Predictive of Elevated Pet Amyloid in Normal Older Adults

Abstract: Early detection and tracking of very subtle cognitive decline due to preclinical AD (e.g., abnormal Aβ on PET imaging) is vital to the success of secondary prevention trials. Here we determine whether more frequent (monthly) assessment of associative memory using a computerized cognitive composite (C3) is associated with amyloid positivity amongst clinically normal (CN) older adults. 76 CN older adults underwent PiB-PET neuroimaging to determine Aβ+/- status (mean age=78.51±5.30, 32% Aβ+) and completed the C3 at-home on a tablet monthly for up to 12 months. The C3 includes the Cogstate Brief Battery, Pattern Separation, and the Face-Name Associative Memory Exam (FNAME). Six-month results for FNAME are reported here. Participants completed two versions of FNAME monthly: one with identical face-name pairs (A-A-A) and one with novel face-name pairs presented (B-C-D-etc). Accuracy of recall for face-name pairs between Aβ+/- over monthly administrations was examined using linear mixed models controlling for age, sex, and education. There were no Aβ+/- performance differences at baseline on face-name memory in either FNAME version. Longitudinal differences between groups were observed for the monthly identical FNAME (Aβ+*Time, β=0.17 se=0.04 p=0.002) but not the monthly novel FNAME (Aβ+*Time, β= −0.02 se=0.04 p=0.602). That is, by the fourth administration of the identical FNAME, Aβ+ CN were benefiting less from repeated exposure to the same Face-Name pairs compared with Aβ- CN who continued to improve with each exposure. By contrast, both Aβ+/- groups performed comparably over time and in relation to each other when they were required to learn new face-name pairs monthly (i.e., novel FNAME).
