Abstract: s from the 19th Annual Meeting 621 4.99 years old) and 15 matched normal controls (mean age 45.01 _+ 7.24 years old) were selected.A comprehensive neuropsychological battery was administered three times over a period of 18 months.Individuals with FAD met the criteria for dementia and were positive for the E280A preseniline 1 mutation.Normal control subjects were the FAD patients' relatives, negative for the mutation. Two groups of neuropsychological instruments were administered: (a) The Consortium to Establish a Registry for Alzheimer's Disease (CERAD) neuropsychological test battery, and (b) additional neuropsychological tests of attention, language, constructional abilities, and memory.FAD patients were significantly impaired on all measures at the first visit except for reading of words and digit forward subtest. While the performance of the normal controls remained unchanged over the 18 months for most neuropsychological tests, the FAD patients displayed a decline in verbal memory, constructional, abstraction, and ideomotor praxis tests. The greatest decline was observed on the Mini-Mental State Exam.Language tests (naming, language comprehension, and verbal fluency) and nonverbal memory defects did not show a further decline and mechanics of reading remained normal over visits. These results support previous findings of the sensitivity of verbal memory tests and the stability of language abilities in autosomal dominant Alzheimer's disease.
