Abstract: A live-cell assay is developed to monitor interhomologue repair of a DNA double-strand break in Schizosaccharomyces pombe, revealing that multiple cycles of homology search and strand invasion occur during repair. It is proposed that these dynamics reflect a preference for repair by synthesis-dependent strand annealing in fission yeast.
